PBMC / Splenocytes isolation

Isolation of peripheral blood mononuclear cells (PBMCs) from whole blood, as well as splenocytes from preclinical specimens, is a critical preparatory step for flow cytometry (FACS)–based immunogenicity assessments, including phenotypic and functional characterization of vaccine‑induced cellular immune responses. VisMederi can provide standardized procedures, operational support, and technical oversight to ensure consistency and data robustness across all study sites. Prior to study initiation, and ahead of laboratory and clinical site training, VisMederi can share its validated PBMC isolation procedure to the sites involved in PBMC collection. Given the highly critical nature of PBMC collection and preservation, VisMederi can supports all stages of the process, including blood sample collection, PBMC isolation by density‑gradient centrifugation (e.g. Ficoll‑Paque Plus using HBSS or equivalent media), cell counting, viability assessment, and long‑term cryopreservation. In addition, VisMederi can design, produce and distribute of PBMC collection kits, including appropriate blood collection tubes (e.g. lithium, heparin or EDTA) and qualified consumables. Following kit distribution, VisMederi can deliver targeted on-site and/or remote trainings to the laboratories involved, with the objective of harmonizing all sites to a single standardized isolation procedure rather than providing basic laboratory training. This VisMederi‑led, standardized approach minimizes inter‑site variability and ensures the reliability, reproducibility, and comparability of FACS‑based immunogenicity data across clinical and preclinical studies.
