Micro-Neutralization ELISA-based (MN-ELISA) assay

The MN‑ELISA is a semi‑quantitative serological assay used to measure virus‑specific neutralizing antibodies and is primarily applied in VisMederi to influenza A and B viruses as well as to a broad range of other medically relevant viruses, including RSV (Establishment and validation of a high-throughput micro-neutralization assay for respiratory syncytial virus (subtypes A and B).), hMPV (Design and validation of a semi-quantitative microneutralization assay for human Metapneumovirus A1 and B1 subtypes ), measles virus, Dengue virus, Zika virus, West Nile virus, HPV, adenoviruses, rotavirus, and hepatitis A and B virus (HAV, HBV). The assay is recommended by the World Health Organization (WHO) and referenced in European Medicines Agency (EMA) guidelines for influenza and other vaccines. It assesses inhibition of viral infection in cell culture following incubation of serially diluted, heat‑inactivated sera with live virus. Viral replication is detected by ELISA using antibodies against viral nucleoprotein or other viral antigens, and neutralizing titers are defined as the reciprocal of the highest serum dilution yielding a signal below a predefined optical density threshold.
