Luminex xMAP
Luminex xMAP technology is a bead‑based, multiplex immunoassay platform that enables the simultaneous qualitative or quantitative detection of multiple analytes within a single serum sample. The assay employs spectrally distinct, color‑coded magnetic microspheres, each coupled to a specific antigen or antibody, allowing concurrent capture of multiple target antibodies or antigens following incubation with the sample. Bound analytes are detected using a fluorescently labelled reporter molecule, and individual bead identities and associated fluorescence signals are measured by a Luminex analyzer using flow cytometry–based detection.
Compared with conventional serological assays that assess single antigen–antibody interactions, Luminex xMAP technology offers substantial advantages, including multiplexing capability, reduced sample volume requirements, increased throughput, and improved workflow efficiency. This platform supports a broad range of applications, including human inflammation panels for comprehensive profiling of inflammatory markers, cytokine panels for detailed characterization of immune responses, human antibody isotyping to assess humoral immune profiles and class switching, customizable singleplex and multiplex bead-based immunoassays for targeted or large-scale biomarker analysis.
The platform enables comprehensive immune profiling against multiple viral and bacterial antigens in parallel, supporting vaccine immunogenicity assessments, sero‑epidemiological studies, and large‑scale clinical investigations, making it particularly suitable for complex study designs and population‑based analyses. At VisMederi, Luminex technology is predominantly used for the assessment of immune responses against bacterial targets (Development and qualification of a multiplexed immunoassay to assess the immunogenicity of Shigella vaccines), while offering flexibility for implementation in viral immunology applications.
