Fluorescent Focus Assay (FFA)

The Fluorescent Focus Assay (FFA) is a quantitative virological method used to determine the infectious virus titer based on the detection of virus‑infected cells by immunofluorescence. The assay measures the formation of discrete fluorescent foci resulting from localized viral replication following infection of susceptible cell monolayers with serial dilutions of a viral sample. After an appropriate incubation period, infected cells are fixed and stained using virus‑specific antibodies conjugated to fluorescent labels, allowing visualization and enumeration of infection foci by fluorescence microscopy or automated imaging systems. Viral titers are calculated and expressed as fluorescent focus‑forming units per milliliter (FFU/mL). FFA is particularly suited for viruses that produce limited, delayed, or no cytopathic effect and is widely applied in vaccine development, antiviral evaluation, and viral characterization for pathogens such as influenza viruses, respiratory syncytial virus (RSV), measles virus, human metapneumovirus, parainfluenza viruses, dengue virus, Zika virus, and other emerging or non‑cytopathic viruses. When appropriately validated, the FFA provides a sensitive, specific, and robust measure of infectious virus suitable for use in regulated laboratory settings.
